چکیده

Sir: As reported previously,1-5) aminoglycoside 6'-N-acetyltransferases [AAC(6')] in R factorcarrying Escherichia coli and Pseudomonas aeruginosa are involved in the mechanism of resistance to several aminoglycoside antibiotics such as kanamycin, kanamycin B, 3',4'dideoxykanamycin B (DKB) and ribostamycin. Gentamicin C1a and sisomicin are acetylated by these enzymes6,7). In this paper, we report the structural requirements of the substrate for the action of AAC(6') prepared from a strain of P. aeruginosa.

Detection of Neuraminidase Activity in Pseudomonas aeruginosa PAO1

Objective(s) Some properties of neuraminidase produced by Pseudomonas aeruginosa PAO1 growth in a defined medium (BHI) were examined and evaluated for its features. Materials and Methods The obtained supernatant enzyme of P. aeruginosa PAO1 cultures was used in a sensitive fluorometric assay by using 2'-(4-methylumbelliferyl) a-D-N acetylneuraminic acid as substrate. As hydrolyzing MUN with ...

Resistance due to aminoglycoside modifying enzymes in Pseudomonas aeruginosa isolates from burns patients.

BACKGROUND & OBJECTIVE Enzymatic modifications of aminoglycosides result in high-level resistance in numerous bacterial species. However, the data on this aspect are elementary in our country. The present study was therefore designed to determine resistance rates and patterns, and to find out the prevalent aminoglycoside modifying enzymes (AMEs) in clinical isolates of Pseudomonas aeruginosa fr...

Aminoglycoside resistance resulting from tight drug binding to an altered aminoglycoside acetyltransferase.

The aacA29b gene, which confers an atypical aminoglycoside resistance pattern to Escherichia coli, was identified on a class 1 integron from a multidrug-resistant isolate of Pseudomonas aeruginosa. On the basis of amino acid sequence homology, it was proposed that the gene encoded a 6'-N-acetyltransferase. The resistance gene was cloned into the pET23a(+) vector, and overexpression conferred hi...

Plasmid-mediated gentamicin resistance of Pseudomonas aeruginosa and its lack of expression in Escherichia coli.

We isolated 11 nonconjugative plasmids mediating resistance to aminoglycoside antibiotics, including gentamicin, from Pseudomonas aeruginosa strains. Their genetic properties were investigated in both P. aeruginosa and Escherichia coli transformants. The plasmid molecular weights ranged from 11 x 10(6) to 24 x 10(6). A low level or complete absence of gentamicin resistance was observed when the...

Pseudomonas aeruginosa 1244 pilin glycosylation: glycan substrate recognition.

The pilin of Pseudomonas aeruginosa 1244 is glycosylated with an oligosaccharide that is structurally identical to the O-antigen repeating unit of this organism. Concordantly, the metabolic source of the pilin glycan is the O-antigen biosynthetic pathway. The present study was conducted to investigate glycan substrate recognition in the 1244 pilin glycosylation reaction. Comparative structural ...
